Real-time PCR for detection of Brucella ovis and Histophilus somni in ovine urine and semen
نویسنده
چکیده
Infectious ovine epididymitis affects rams worldwide, and it is considered to be one of the most important infectious diseases (Poester et al., 2013). Brucella ovis is one of the most important infectious agents associated with this disease, which may also be due to Histophilus somni infection, as well as other organisms (DíazAparicio et al., 2009; Poester et al., 2013; Moustacas et al., 2014). B. ovis infection is associated with chronic epididymitis and infertility in rams and, occasionally, abortion in ewes and birth of weak lambs. H. somni is a component of the microbiota colonizing mucosal surfaces of cattle, goats, and sheep. However, H. somni may act as an opportunistic pathogen, and the infection may result in epididymitis as well as several other clinical manifestations such as vaginitis, placentitis, pneumonia, meningoencephalitis, mastitis, synovitis, septicemia, and other reproductive disorders (Díaz-Aparicio et al., 2009; Poester et al., 2013; Moustacas et al., 2014). The gold standard diagnosis of B. ovis infection is based on clinical examination, serology and semen bacteriology (Xavier et al, 2011; Poester et al., 2013), while for H. somni infections, only clinical examination and bacteriology are routinely performed. Semen and urine are the samples of choice for diagnosis (Xavier et al., 2010; Costa el al., 2012; Moustacas et al., 2013). These techniques are labor intensive and slow, whereas molecular techniques have been increasingly used, including conventional PCR (Xavier et al.,

Species-specific multiplex PCR for the diagnosis of Brucella ovis, Actinobacillus seminis, and Histophilus somni infection in rams
BACKGROUND Infectious ovine epididymitis results in substantial economic losses worldwide due to reproductive failure and culling of breeders. The most common causative agents of these infections are Brucella ovis, Actinobacillus seminis, and Histophilus somni. The aim of this study was to develop a multiplex PCR assay for simultaneous detection of Brucella ovis, Actinobacillus seminis, and His...
متن کاملSpecies-specific nested PCR as a diagnostic tool for Brucella ovis infection in rams
The aim of the present study was to evaluate a species-specific nested PCR based on a previously described species-specific PCR for detection of B. ovis in semen and urine samples of experimentally infected rams. The performance of the species-specific nested PCR was compared with the results of a genus-specific PCR. Fourteen rams were experimentally infected with the Brucella ovis REO 198 stra...
متن کاملDifferentiation between bovine and ovine strains of Histophilus somni based on the sequences of 16S rDNA and rpoB gene.
Nucleotide sequences of 16S rDNA and rpoB gene of 25 bovine and 6 ovine Histophilus somni strains were determined to detect subtle differences between the host animal species. The 1465 nucleotide residues of the 16S rDNA exhibited levels of sequence similarities of 99.4% or more. The high sequence similarity of the 16S rDNA of recently described species H. somni was confirmed in the 31 strains ...
متن کاملEvaluation of a new set of Real-Time PCR for Brucella detection within human and animal samples
A quantitative TaqMn Real-Time PCR assay was developed and its diagnostic value on human serum and livestock samples were evaluated. Brucella species could be distributed through communities as a biological agent. Rapid detection of biological threat agents is critical for timely therapeutic administration. Quantitative real-time PCR provides a rapid, sensitive and specific tool for molecular i...
متن کاملDevelopment of New Modified Simple Polymerase Chain Reaction and Real-time Polymerase Chain Reaction for the Identification of Iranian Brucella abortus Strains
Brucellosis is primarily a worldwide zoonotic disease caused by Brucella species. The genus Brucella contains highly infectious species that are classified as biological threat agents. In this regard, the identification of Brucella can be a time-consuming and labor-intensive process posing a real risk of laboratory-acquired infection to the laboratory staff. This stud...
